The soybean aphid (Aphis glycines Matsumura ) is one of the main pests of seedling stage of soybean, because soybean aphid produce resistance to chemical pesticide, people enhance consciousness to environmental protection at the same time, the biological control of the soybean aphid is more and more attention to by people. Beans contain abundant natural enemy insect resource in soybean fields, how to utilize reasonably a large number of valuable natural enemy resource has already become one of the research focuses in biological control of soybean aphid.The research method of predator-prey interactions is direct observation or immunological assays, but direct observation is apt to bring the error, immunological assays is more expensive. the molecule technology based on DNA especially PCR technology have already been applied to research predator-prey interactions with the wide application of biotechnology in entomology research. mtDNA has advantages over nuclear DNA in estimating Mitochondrial DNA has advantages over nuclear DNA in estimating phylogenetic relationships among insect taxa because of its rapid evolution and inheritance without gene recombinations,so mtDNA is widely used in study on molecular. This experiment cloned and determined cycochrome oxidaseâ…¡(COâ…¡) gene sequence of soybean aphid, and analysis gene sequence of COâ…¡, the relationship between predator and soybean aphid were evaluated by using PCR.The main results of the study as follow:Cytochrome oxidase subunitâ…¡gene segment(registration No.DQ265743) of soybean aphid was cloned and sequenced, the sequence was sented to GeneBank with registration No.DQ265743. Aphis glycines was compared with Rhopalosiphum padi, Sitobion avenae, Schizaphis graminum, Rhopalosiphum maidis, Sipha flava, Diuraphis noxia, Aphis gossypii, Myzus persicae, The homology of the COâ…¡gene nucleotide sequence is 92.7%, 92.8%, 91.5%, 93.0%, 89.1%, 91.9%, 95.6%, 93.2% respectively, amino acid sequence is 97.1%, 96.1%, 96.1%, 97.1%, 89.7%, 95.1%, 97.8%, 95.4% respectively.The nucleotides composition of COâ…¡gene from soybean aphid was analyzed, the results showed that the content of A, T, G, C are 40.43%, 39.01%, 8.37%, 12.20% respectively, and A +T content is 79.43%, G+C content is 20.57%, the content of A+T up to 94.04% on the third site of codon, which shows a strong A+T bias.The COâ…¡gene 672 bp sequence of soybean aphid coding 223 amino acids, the frequency of codon with A,T ending is high, has no codon with G eding.In the composition of amino acid, the content of Leu is the highest, and is 18.39% of the total amount, the content of Ala, Cys, Val is the...
