Two F2 populations developed from cold type cultivar ('RB6','xiaoyan6') and warm ones ('9430','NR9405') are produced. The distribution characters of flag leaf temperature (FLT) during reproductive growth stage were revealed. The correlation between FLT and varied flag leaf structure characters (leaf thickness, transverse section area of vascular bundles and interval between pairs of adjacent bundles) and the correlation between FLT and number of green leaves (NGL) were studied. Finally, the effects of FLT and NGL on 100-kernel weight were analyzed in the two F2 populations. The results prove that the FLT highly correlates with transverse-section area of vascular bundles and the leaf thickness, while not do with density of vascular bundles. 100-hundred kernel of individual is greatly affected by FLT and NGL during later reproductive growth stage.In order to improve the efficiency of RAPD analyses, this research illustrate the distribution character of the number of combination locus and the number of amplification products. Computer programs were designed to simulated PCR reactions of RAPD in some genome sequences of model organisms. The results prove that primer bias exists in DNA sequences and that the biases of different organisms do not coincide with each other. And then, through regression analysis the linear regression equation about the number of amplification products and the number of combination locus is built. The right of this regression equation is tested using simulation method. It is proved that primers selection making use of bio-sequence information will improve efficiency remarkably.Optimal reaction conditions for RAPD analysis are established. It was finally determined with 25-μl reactions containing: 10ng of plant genomic DNA, 2.0mM Mg2+, 0.24mM primer and 100-400mM of each dNTP and 1.0 units of Taq DNA polymerase. Two electrophoresis methods for amplification products are compared. The results illustrated that 5% PAGE has a higher resolution than 1.8% agarose.
