| The research was carried out on the study of the compatibility between Cardiucrineum giganteum and Lilium, the in vitro tissue culture of C . giganteum and its tolerance under heat stress,in order to provide evidence for the Cardiucrineum breeding, germplasm storage and the multiplication and cultivation of its new cultivars. The main results are showed bellow:The pollen tube behavior in the style following the hybridization between Cardiocrinum giganteum and Lilium were observed by means of fluorescence microscopy. The results indicated that the pollen of Lilium could germinate on the stigma of C.gaganteum and the pollen tubes could reach the basal part of the style and finally arrived at the ovule in ovary, meanwhile, the result also showed that the speed of the pollen germination and pollen tube elongation following hybridization were much slower than that following intraspecific pollination. Several abnormalities of pollen tubes were observed including pollen tubes with branching off , swollen or pointed tips, or superposition in the middle parts lots of callose depositiing at irregular distances, parts of pollen tubes became wave-liked at the middle or basal part of the style, which finally led to the failure to arrive at the ovary.The bulb scales, leafstalk and ovary of C.giganteum were used as explants for in vitro tissue culture.
