Pulmonary Hypertension Syndrome of broiler(PHS) that has a group of character symptom such as ascites and heart failure, is also one of major diseases threatening broiler industry. Endothelin-1 (ET-1) is a potent vasoconstrictive peptide that was isolated initially from the conditioned medium of cultured endothelial cells. It is a pathogenic mediator with a number of deleterious effects, including vasoconstriction, fibrosis, vascular hypertrophy, and inflammation.Atrial natriuretic peptide, angiotensin II and nitric oxide also takes an important role in cardiovascular system. The tests, by laboratory conditions to cultivate pulmonary artery endothelial cell of Broilers, to obtain the test cell required, and then through the granting of PHS's different concentrations ET-1 and L-Arg factors, L-Arg + ET-1 and L-NAME to observe the impact of cell secretion. The test results show that through the adoption of foreign origin ET-1 incentives cell secretion NO, ANP and AII ET-1 concentrations increase with the volume of the increase, and the role of the endodermis cells are promoting multiplication, a concentration dependence. These changes are NOS cNOS and iNOS reduce the increase, resulting in a general rise NOS promote the secretion increased NO. Fall outside the L-Arg complement to the promotion of pulmonary endodermis cell secretion no increase, but no impact on cNOS and iNOS. NOS inhibitors reduce cNOS L-NAME through the reactive for the suppression of pulmonary endodermis cell NO generation, demonstrated by its strong cNOS selective. L-Arg + ET-1 common role injury endodermis cells, the biological effects are ET-1 play a major role as the lead role, thus affecting pulmonary endodermis cell secretion change. Through this study, revealed in the normal biological intimate represented by ET-1 AII vascular factors such as number and no represented ANP, dilate factors common in cardiopulmonary system synergies, the maintenance of normal body balance in Broiler ascites syndrome occurred development process plays an important physiological and pathological roles.
