The Establishment And Primary Application Of A Fast Diagnostuc Method Detecting Reticuloendotheliosis Virus

Reticuloendotheliosis(RE) is a tumorous disease of turkey, chicken, duck, partridge, pheasant, goose caused by reticuloendotheliosis virus(REV), which is characteristic of hyperplasia of lymphocytic netted cell. The lethal function of REV is not obvious to birds , but REV can cause the bursa to wither, its pathogenesis gene V - rel can cause the B lymphocyte to transform as the tumor cell, damage the birds organism immunity system, and cause organism resistivity to drop, thus the gene can cause other concurrent diseases and create a high elimination rate and a high mortality rate; The serology investigation indicated that the ratio of the chickens infected REV is very high in our country. So, it is urgent to establish a method to detect and control this disease.The procedure of capture ELISA for detection of REV antibodies were established with the monoclonal antibodies 11B118 against REV, serum-anti chicken and purified antigen. The results revealed that the variation coefficient of the ELISA plate is 2.9% and it is up to the ELISA condition; the optimal concentration of monoclonal antibody is 1:1280; the optimal closed liquid is 1% BSA and its optimal woking time is 37℃1h; the dilution coefficient of antigen and antibody is 1:320 and 1:20 respectively; the working concentration of HRP-labeled rabit anti-chicken Ig is 1:15000;the woking time of the serum and HRP-labeled rabit anti-chicken Ig is 37℃ 1h;the reaction time of TMB is 10min at room temperature. According to the the procedure of capture ELISA, the critical value of S/P is 0.253. 200 serum samples were detected by the self-ELISA and IDXX-ELISA respectively, the accordant rate is 87%.
