In this experiment, we used cotyledon of all the 26 lines of cucumber (Cucumis sativus L.) as the materials for genotype selecting. There are 6 lines could be regenerated in a high rate. Than we studied the effect of PPT on these 6 lines to confirm the concentrations in shoot induction media. We used cotyledon of S52 (FF) to be the explants, optimized shoot induction media and tested the PPT concentrations in rooting media. S52 was transformed with Agrobacterium tumefacien LBA4404 as the mediator that carries the binary vector pEZT-NL containing an ACS1 gene and bar gene. It is looking forward to study the effect of ACS1 gene in influencing the flower sexual expression.The 26 cucumber lines are 6 South China types(ff): S50, S52, S53, S54, S78, S85; 6 North China types(FF): S61(ff), S68(ff), S69(ff), S73(FF), S92(ff), S94(ff) and 14 European types: S02, S04, S07, S17, S43,...
