Studies on plant salt tolerance has been a research interests for the past decade focused mainly on annual herb plants as Arabidopsis , Thellungiella Salsuginea as well as a great deal of agricultural crop, but comparatively less in perennial woody plants. Populus eupharatica is a valuable salt-tolarant tree specices distributed in China's Northwest dry and saline district, and the only large tree species which is able to survive in the deserts in Xin Jiang Autonomous Region.of China. And it has making a unreplaceable contribution to the fixing of moving dune and building of the network of the farm or woods. Salt-tolerant mechanism of Populus eupharatica is becoming a research focus with its strong salt tolerance. There were some scientific reports focused on salt-tolerant mechanism of the plant in the aspects of plant physiology in last years, but the molecular mechanism of why it can tolerate salt stress is still unclear. So It is significant to explore the molecule mechanism and isolate the genes involved in salt-tolerance of populus eupharatica. Through this kind of stuies, the foundations for applying the genes of Populus eupharatica in breeding new salt-tolerant trees will be madeIn this paper,Affymetrix GeneChip Poplar Genome Array was used in studing gene expression profile of Populus eupharatica by using leaves of the salt treated seedlings and the leaves of seedlings without any treatment In this experiments RNA preparation chip hybridization, chip image detection and data-processing was conducted. And 1102 differencailly expressed genes was observed. Among these differencially expressed genes 454 genes were up- regulated in respone to salt stress and 558 genes were down-regulated. These differencially expressed genes involves in different biological process as ion transport, osmoregulation, signal transduction ,growth and development. The result of the presented studies will make some contributions to the understanding of salt-tolerant mechanism and meanwhile make a base for further applying the genes of Populus eupharatica in breeding salt tolerance trees.
