Rapeseed is one of the most important oil crops, and also is one of the major economic crops. It occupies an important place in our national economy. Its oil is for cook, and the meal is for feeding the animal In recent years, the oil quality is continually refined and gradually applied in chemical industry, energy industry, and etc. Genetic modified rapeseed is to produce the raw material of biodiesel. As to the application of rapeseed continually widen, the research on it has been intensifted either. It is become the most important goal that modifying the contents of oil by gene engineering technology to improve its economic properties in recent years.Phosphoenolpyruvate carboxylase (PEP) plays an important role in the control of the ratio of the protein/lipid content in rape (Brassica napus).To modify the expression of PEP gene in rape by co-suppression may enhance the oil content of the transgenic plants. In this research, a PEP gene fragment was amplified from Huayou5(H045) with polymerase chain reaction(PCR) and then cloned into the corresponding sites of the seed-specific expression vector pBI121-B in the antisense orientation. Antisense PEP gene was introduced into Huayou5(H045) successfully Via laser microbeam puncture, and the transgenic plants which were confirmed by kanamycin resistance screening and polymerase chain reaction(PCR) analysis had been obtained. In this research the foreign gene was introduced into rape successfully Via laser microbeam puncture, which avoids the problems of Agrobacerium tumefaciens transformation, such as the low frequency of transformation, difficulties of repetitive, the limitation of genotype and so on. It provides a new way to enhance the oil content of the seeds of rape through the new genetic engineering technology, and has enriched the rape high oil breeding theory.
