High grain-protein-content(GPC) wheat varieties have been obtained by ion beam-induced mutation and ion beam-mediated transformation in our lab. Their GPC were increased by 3% to 5% compaired with CK and the highest GPC surpassed 20%. It isn't well known that the reasons of GPC improvement and the relationship between GPC and wheat qulity traits, although these wheat varieties have stably inherited to the fifth generation. Storage protein is the main constituent of wheat grain protein, and it mainly includes glutenin and glidian, respectively accounts for approximately 40% of seed protein. Glutenin is also composed of high-molecular-weight glutenin subunits (HMW-GS) and low-molecular-weight glutenin subunits (LMW-GS) according to their mobility in sodium dodecyl sulphate-polyacrylamide gel eletrophoresis (SDS-PAGE), and numerous studies from home to foreign indicate that the glutenin has spacious effects on wheat quality. Therefore, this research analyses the variance of HMW-GS and glidian respectively by the SDS-PAGE and A-PAGE.SDS-PAGE experiment conditions suitable for analysing HMW-GS variation wheat were established by taking the Yumai 52 as the material, after the optimization of five limiting factors in SDS-PAGE selective electrophoresis: concentration of gel, activator, voltage, the sample buffer, and the sample quantity. HMW-GS constituent of Yumai 52 is 1,7+9 and 5+12 subunits determined by optimized SDS-PAGE. It indicates that HMW-GS of 74 samples of high grain-protein-content wheat varieties have remarkably increased the expression of grain protein without different bands by optimized SDS-PAGE. In addition, in order to achieve clear electrophoresis bands,we also optimized the experiment conditions firstly from five limiting factors in A-PAGE: concentration of gel, activator, voltage, the sample buffer, and the sample quantity. In the result, the expression quantity of grain protein has increased without different bands compaired with CK inα,β,γ- glidian, but 40 samples all increase two bands compaired with CK inωsection, the rates of different bands is 54.05%.The results show that the average GPC of high grain-protein-content wheat varieties and their the rates of different bands are impositive correlated .The GPC and SDS sedimentation value are all very important indexes which can evaluate wheat quality, therefore this research has measurated the SDS sedimentation value of high grain-protein-content wheat varieties and analyzed the relationship between their SDS sedimentation value and average GPC. In addition, the contributions of each HMW-GS to SDS sedimentation value of high grain-protein-content wheat varieties were also studied. As a result, the SDS sedimentation value of high grain-protein-content wheat varieties were higher than that of CK and that are also positive correlated to their average GPC.
