Preparation of transfer factor nanometer liposome and detection of efficiency of antivirus, offer a new prevention and method for viral disease and solve problems of animal food safety.Nonspecific transfer factor (TF) was prepared by dialysis method from the spleens of healthy AA broiler. TF solution was then made into powder by lyophilization, and was diluted to detect the physicochemical property. The protein in TF was negative reaction. Content of polypeptide exceeded 0.1mg/ml. OD260/OD280 value was 2.39. The amino acids were 17 species in TF. The results of detection were identified with document.The transfer factor nanometer liposomes (TFNL) were prepared by reverse evaporation technique. Using encapsulation efficiency as index of examination and designing an orthogonal experiment, the optimal formulation of was determined. Morphology character and confluence were observed under electron microscope. Detect encapsulation efficiency and percolation rate in different time. The optimal preparation formulation: PC/CH(molar ratio):2:1, TF concentration (TF): 60mg/ml, Tween-80:1ml, oil-phase/water-phase (volume ratio):4:1. TFNL grain size was less than 100nm and distributed symmetrically. Average entrapment efficiency was up to 49.5%. As the time gone, TFNL particle was confluent and percolation rate was augmented, but the velocity of permeability did not speed up. Four and half months later the rate of leakage was about 52.4% The result manifested that liposomes were stabile and expiration was 4.5 months.Detecting the safety, anaphylaxis of TFNL by the effect on cellular immunity,and using erythrocyte rosette formation test and macrophage phagoytosis test. The results suggested that: TFNL was asepsis and promoted immune function.Observe the small intestine of mice orally under light microscope and electron microscope. The result was that lipid droplet appeared in experimental group mice intestinal villi, which suggested that liposomes peridium TF could avoid TF degraded by gastric acid enzyme and absorbed well in small intestine.ED50 of TFNL was determined by Karber's method. Anti-virus experiment: Chickens were immuned with taken TFNL orally, and then inoculated NDV at seventh day. Relevant items for instance clinical symptom, antibody, and mortality. Pathological changes under microscope were recorded. The results were: ED50 was 0.07mg/ml. Medication concentration was 0.14mg/ml. Anti-virus effect: Every experimental groups had processed different effect of anti-virus, and TFNL used with vaccine was the best.TFNL treated REV, and the TFNL had energetic activity, but the chickens at death end. The results suggested that TFNL had certain therapeutic action but no therapeutic value.This foundation indicated that TFNL was asepsis and has no side effect. Expiration was 4.5 months. Absorbed well in small intestine,TFNL as adjuvant had processed different effect of anti-virus and had certain therapeutic action. So TFNL as vaccine adjuvant has enormous generalization value in production industry.
