| Drought and salt is the serious problem all over the world, which is also the important factor of in agriculture producing in our country .Xingjiang belongs to drought and desert area, where there are all kings of heavy salt areas. Meanwhile , Xingjiang is one of the biggest cotton basic areas, the fields which are opened up wasteland and planted increase, but the cotton needs lots of water , the crisis of using water becomes remarkably. Drought and salt restrict the improvement of cotton industy in Xingjiang. For many years , the persons solve the problem of drought and salt by using traditional dreeding methods ,but it costs much money and needs a great deal of water, the efficiency of selecting is not high. AS the biological developing, breeding the cotton of resisting drought and salt will be possible. some scholars study and improve differential display reverse transcription PCR( DDRT-PCR), which is used to clone the genes resisting adversity and obtain the new transformed cotton. At present,the gene of rice resisted salt,wheat springed,rice resisted seasonal febrile diseases are separated by using DDRT-PCR.The cotton resisted drought and salt which normally grow and are dealed with different concentration PEG6000 and NaCl are considered as the material of study.A physiological study on cotton in drought and sait stress,and identify the 120 kinds of cotton of drought and salt tolerance,analyse their relations.Different fragments are obtained through the method of DDRT-PCR, then the fragments are identified and analyzed with the results as follows:1. the 120 kinds of cotton of drought and salt tolerance are evaluated .The biggest reduced concentration of PEG60 and NaCl are 25% and 1.3% for choosing the cotton resist . Drought and salt Zhong07,C6015,KK-1543,NongKen5 are considered as the material highly resisted drought ZhongR2081,ZhongR2080,ZhongR2069,SuK202(land-cotton) are considered as the material highly resisted salt. C4744,C1470,XingLuzao1 are considered as the material which are hardly resisted drought and salt. C6020 and Kui85-174 are considered as the material which are resisted not only drought but also salt.2. Different physiology influence on cotton are separately identified before inducing,inducing after 1.5 hours and after 3 hours,but inducing after 1.5 hours , different physiology of cotton change greatly. Different index are evaluated synthetically, index measured reflect cotton of drought and salt tolerance.3. mRNA differential display system of cotton is established for choosing the cotton resist . Drought and salt. The 240 combinations which are established by 12 anchored primers and 20 arbitrary decamers are conducted by RT-PCR,the products are applied the method of silver-staining by 6% PAGE. 88 differentially displayed fragments are obtained, and 41 of them which display stably are picked up by the second PCR., by both analysis of nucleotide and deduced amino acid sequence similarity, 7 sequences are obtained with high identity more than 85%,which show sequence obtained are related to resisting drought and salt. |
