Hydatid disease (HD) is a severe zoonosis which caused by lavar stage of Echinococcus, It results in public health and economical problems in the world. The dogs as a definitive host play an important role in transmission. It is a pivot that we control familys or herds dogs on prevalence. we directly vaccine the dogs to prevent eggs from transmission that it could control infectious origination and protect the intermediate host infect HD. So, it is very effective and feasible to vaccine dogs to cutting the cycle of HD. Zhang et al used to test differences in mRNA expression between immature adult worms (IAW) and mature adult worms (MAW) of Echinococcus granulosus were determined using polymerase chain reaction-based differential display (DDRT-PCR) and choose the special genes of EgM family . The EgM9 of EgM could effectively express in E.coil and purify.The test detected antibody by ELISA and cytokine be used Real Time PCR which vaccined for dogs EgM9 protein combinant adjuvant QuilA.We got the results the IgG dominant antibody and its isotype subclass existed distinctly different responses between vaccination group and control group. Also, the interferon-γ(IFN-γ)in 15th week (w) is as more1000 times as in 0w ,while the IL-4 in 15th week is as approximate 20 times as in 0w.The results showed that EgM9 recombinant protein could produce Th1 and Th2 response , which activated the humoral immune and cell-mediated immunity response.After be challenged protoscoleces (PSCs) dogs were necropsied on 45 days.It is found that the EgM9 group had average 7573 worms only possession 3.79% and 3060 gravid proglottids only possession 1.5% in infectious Protoscoleces , but the control group has average 57080 worms reached 28.5% and 21900 gravid proglottids reaching 11.0% in infectious Protoscoleces. The results showed the EgM9 group,s decreased eggs 86% ,contrasted with control group. This test proved EgM9 recombinant protein vaccinated the dogs could obtain the obviously protective efficiency.
