The study was tested two culture medium (NCSU-23 and PZM-3), results showed thatPZM-3 was superior to NCSU-23 for the development of hatched stage of parthenogeneticactivation (PA) porcine embryos, the hatched rate is significant difference between them(32.6% VS 18.9%, P<0.05). Amino acid is contained by PZM-3 but not NCSU-23. Thesubsequent experiment showed that, with NCSU-23 supplemented, addition of 2% BEMessential amino acid (EAA) solution significantly decreased the PA embryo formation(20.1% VS 25.1%, P<0.05), addition of 1% MEM Non-essential amino acid (NEAA)solution resulted in a significant increase in embryo formation(24.7% VS 19.7%, P<0.05),however, the addition of both EAA and NEAA, data were not statistically different amongtreatments.In the second part, the effects of four antioxidant additions (ethylenediaminetetraaceticacid (EDTA),β-Mercaptoethanol (β-ME), vitamine E, superoxide dismutase(SOD) on thePA procine embryo development were investigated. The results showed: addition of100μmol/L EDTA significantly increased the embryo formation (24.1% vs 18.0%, P<0.05),the same to the treatment for addition of 50μmol/Lβ-ME (22.8% VS 19.1%, P<0.05).Then,among treatments the addition of Vitamine E data were not statistically different(P>0.05). However, a dramatically reduction on the cleavage rate and embryo formation(56.0% VS 64.7%, 9.7% VS 16.3%, P<0.05) after addition of 300IU SOD.In conclusions, (1) PZM-3 was better than NCSU-23 in hatch of porcine PA embryos.(2)With NCSU-23 supplemented, 1% NEAA is benefit to porcine PA embryo development, but2% EAA is detrimental. (3) Addition of either 100μmol/L EDTA or 50μmol/Lβ-ME isbeneficial to the development of porcine PA embryo, (4) Vitamine E may promote the porcinePA embryo development, which will need to be confirmed further. (5) SOD is detrimental tothe porcine PA embryo development.
