This experiment adopt four adjuvant (APS,Propolis,FIA,Al(OH)3Colloid) with LaSota lived vaccine were carried out in chickens in attemptto analyze the adjuvant effect. The effect after immune LaSota lived vaccinewere evaluated for their ability to activate B lymphocytes to induce HIantibody, promote T lymphocytes proliferation, induce macrophage nitricoxide (NO) production, protection rate of from NDV challenge and growthafter of chickens in this study.Result after counts software, showing APS group a significantincrement from the control group (inoculated with Lasota lived vaccineonly) (P<0.01); APS group compared with Propolis's (NO is not),Freund's Adjuvant (FA),Al(OH)3 group are difference(P<0.05).The forth weeks after immu, four adjuvants' target are all highest inbirds, four adjuvant presented respectively mean serum HI antibody titerof 8.8,8.6,7.8,8.2 log2, lymphoproliferative stimulation index of 5.39, 4.94,3.25,3.71, macrophage NO production of 23.82,24.25,17.82,18.32umol/L, protection rate of 100%,90%,80%,90%,from NDV challengewere observed; It demonstrate APS are able to notably enhance specificantibody responses and stimulate a strong cellular immune response toLasota live vaccine on San-huang chickens.Test research the best work density further. APS (100μg/ml),(200μg/ml) and (400μg/ml) with Lasota lived vaccine were carried out.Result after counts software, shows that 200μg/ml group a significantincrement from the control group, 200μg/ml group compared with100μg/ml is difference(P<0.05). 200μg/ml group is high compare to400μg/ml. The forth weeks after immu, three group presentedrespectively mean serum HI antibody titer of 8.6,8.8,8.8log2,lymphoproliferative stimulation index of 5.10,5.59,5.37, macrophage NOproduction of 24.75,26.18,25.75umol/L, protection rate of 100%,100%,100% from NDV challenge were observed. It demonstrate 200μg/mlgroup is best among three group, can enhance specific antibody responsesand stimulate a strong cellular immune response to Lasota live vaccine onSan-huang chickens.In conclusion, data reveal that APS is able to significantly enhanceimmune responses to LaSota live vaccine against ND in Guangxi San-huangchickens, may be exploited as vaccine adjuvant further. 200μg/ml is notableimmune enhancement effects, and has more superiority; It is the best work density, has very big application prospect. These results will be helpful forunderstanding application of adjuvant in poultry ND vaccine, andtheoretically provide an approach for development of novel adjuvant forrcompound vaccine. Test cell immune' way in this experiment, will providean approach to test cell immune.
