Chromosome number and karyotype of Rosa multiflora Thunb. var.cathayensis Rehd. were analyzed. The result showed that the Chromosome was triploid. The karytoye was 2n=3x=21=18m+3sm , which belonged to"2A"type . Two methods were used for genomic DNA isolation of Rosa multiflora and rose. The results indicated that CTAB(accede to steel ball)was the most suitable one. Via estabishment and optimization of PCR technical system in Rosa multiflora, Ten kinds of SSR primers could produce polymorphic bands from eighty-eight kinds of SSR primers.The rotio of polymorphic band is 47.62%. By using five primers in Rosa multiflora and rose, these SSR data were analyzed using cluster methods of UPGMA in the software package NTSYSpc. Thirty-two trials was divided into four cluster groups, respectively. While R . multiflora and Wild Roses belonged to one group. 'Rosy Carpet'and'Hula Girl'were belonged to one group. Meanwhile ,'Timly Snow','Korea','New Flor 1','Royal Basssino'were belonged to one group, and the others were one group. Crossbreeding was done while formulated breeding strategy for improving R . multiflora and selecting horticulture breed. The result validated the feasibility of SSR marker in crossbreeding of R . multiflora Thunb. and Modern Rose Hybrid. Eight F1 hybrids was electd by observing thire character and adapt capability. By SSR molecular marker the hybrid F1 of R . multiflora and'Brocade Cold 1'were dentificated. Five trials was divided into two cluster groups, respectively. While hybrid 5-2, hybrid 5-4 and R . multiflora belonged to one group. Hybrid 5-1, hybrid 5-3, hybrid 5-6 and'Brocade Cold 1'were belonged to one group.It is same with morphology.
