Maize is one of the main crop plants in our country. Following the improvement of the society and the change of the environment, the production of maize is more and more restricted by the stress such as salt, drought, low temperature and crop pest etc. Most of maize species in our country show low tolerance to stress. So it is essential to breed new maize species that have excellent character of stress tolerance.By now, the traditional breeding research in our country had improved slowly and there was rarely good species in the past years. By the rapid improvement of transgenic technology, the genetic engineering assisted breeding has achieved success which traditional breeding couldn't get. So it is an effective approach to use genetic engineering to gain stress tolerance relative genes, breed good crop species and finally improve the situation of our maize breeding and production.Huang Zao Si is regarded as the best inbred line in our breeding domain. It has been widely used for its high and stable combining ability, upright-leaf plant pattern and elite agronomical characters. As a parent, it has the most hybrids number and the largest spred area. Huang Zao Si has made great contribution to maize breeding and production in China.We constructed a bacterial artificial chromosome (BAC) library in this research based on the genome of maize Huang Zao Si which is a good inbred line. This library contains about 153,600 clones which are stored by single clone style. Identification of 300 random recombinants indicated that the average insert size is 92kb, more than 98% recombinants and less than 2% empty clones. The library is steady and has few organelles pollution. It represents approximately 5.6 genome equivalents. Based on the research, we screened the library by stress tolerance relative probes and got some clones which may contain object genes. This result requires further research.Through the construction of the BAC library of Maize Huang Zao Si, this research builds a technique platform for maize genome research and screening of stress tolerance genes. And based on the library, we made a primary exploer for stress tolerance gene. This is very important for maize breeding and production of our country.
