The taxonomy position of Microsporidia cause intense argument in the world.Here, cloned partialα-Tubulin,β-Tubulin gene of Nosema bombycis(Zhejiang),and used Bioeditor software translate nucleotide to amino acids.Another,collected severalα-Tubulin,β-Tubulin genes of different species publiced in GenBank from NCBI.Used CLUSTALX(1.81),Mega2 softwares and another online software CLUSTALW(1.83)analysis sequences and construct phylogenetic trees. The results indicated that the gene with size 1202bp has 99% homology compare toα-Tubulin of Nosema bombycis(Taiwan),and have five nucleotide mutation,three of them near 5'cause three amino acids change; 96%~98% compare to other species of Nosema genus, the sequence had submit to GenBank(Assension No.EF051590);similarity, the gene with the size 1161bp gene has 98% homology compare toβ-Tubulin of Nosema bombycis(Taiwan),and have fourteen nucleotide mutation,four nucleotide mutation near 3'cause four amino acids change; 96%~98% compare to other species of Nosema genus,the sequence had submit to GenBank(Assension No.EF151928). Based on sequence homology analysis,it found that these two genes have different mutation rate,β-Tubulin is faster thanα-Tubulin.The results of phylogenetic analysis indicated that use the same methods with different genes or use different methods with same genes lead to different conclusion.In this reseach ,it gain concordant conclusion is that Microsporidia as a single group is a member of fungi,and has nearest relationship to Entomophaga and Conidiobolus genus of Zygomycetes byα-tubulin andαβ-tubulin phylogenetic analysis; otherwise, Microsporidia has nearest relationship to Basidiomycetes and Chytrids byβ-Tubulin phylogenetic analysis.So,by this reseach ,it can not gain more practice and penetrating conclusion now.In future, it should set a standard system and method to do organism evolution and taxonomy reseach,in another,it should select several genes which participate and represent different functions and characters to gain synthetic analysis.
