To develop artificial insemination technique between male Muscovy duck and female domestic duck, series of experiments were conducted with male Muscovy duck and female Cherry Valley duck, to study procedure of artificial insemination technique. 30 male Muscovy be trained for collecting semen with method of massage or the combination of massage and female duck stimulate and Insemination the female duck with vagina-insemination method. These series of experiments were designed to investigate effects of volume of semen, insemination interval, insemination time and diluent on fertilization rate. The main results were as follows:1.0.89±0.13ml semen was collected from male Muscovy duck with Massage method and the sperm concentration was (1.85±0.19)×10~9/ml, sperm motility was 8.71±0.45 and pH of fresh semen was 7.20±0.13. With the combination of massage and female duck stimulate, semen volume collected was 1.03±0.14ml and sperm concentration was(1.88±0.14)×10~9/ml, sperm motility was 8.82±0.58 and pH of fresh semen was 7.22±0.15.2. The average fertilization rate was only 39.58%by natural mating between male Muscovy duck and female domestic duck, while the average fertilization rate increased to 74.79%by artificial insemination with insemination time between nine and ten in the morning, 0.05 ml semen per duck and insemination interval of three to four days. Fresh semen collected should be used in 30-40min.3. Compared with natural mating, artificial insemination practice make utilization ratio of male Muscovy duck increased ten times and more mule duck can be produced with less cost. 4. The effects that one share of fresh semen was diluted with two share of different diluents (0.9%physiological saline, egg yolk & dextrose solution, phosphate buffer, Lake's diluent) were investigated. Compared to fresh semen of which the fertilization rate was 74.10%, that the fertilization rate of diluted semen by phosphate buffer and Lake's was 75.24%and 75.16%individually (P>0.05). When insemination was carried out after diluted semen stored at 2℃to 5℃for 24h, the average fertilization rate of these two diluted semen decreased to 23.76%and 34.33%individually..
