| Swine chlamydiosis, caused by Chlamydia psittaci, is an important infectious disease,manifesting with abortion, still birth, testis inflammation, pneumonia, enteritis, arthritis andso on. In the study, 1213 sera specimens sampled from pigs in different districts weremeasured for antibody levels to chlamydiosis by means of indirect hemoagglutination (IHA)test. The test results showed that the detectable rates of chlamysis positive antibody were22.68ï¼…in Nanin, 16.39ï¼…in Yulin, 22.27ï¼…in Wuzhou, 28.70ï¼…in hezhou, 18ï¼…in Guigang,26.36ï¼…in Liaozhou, 22.35ï¼…in Laibin, 10.29ï¼…in Guilin, 13.43ï¼…in Beihai, respetively.The detectable rates of chlamysis positive antibody in the pigs of various ages weredifferent: piglets 16.89ï¼…, breedersaws 19.71ï¼…, finishing pigs 21.97ï¼…, respectively. Thedetectable rates of chlamysis positive antibody in the pigs of different species were asfollows: Landrace 20.44ï¼…, Large White 20.07ï¼…, Duroc 19.29ï¼…, Hybridzation 22.19ï¼…,respectively. 386 sera specimens sampled from 21 pig farms with abortion symptom weredetected, out of which 14 farms turned out to be positive (14/21) and 197 sera specimenspositive(197/386). The results showed that the swine chlamydiosis prevalence in Guangxiprovince was serious.Based on the gene sequence of main outer membrane protein (MOMP) of Chlamydiapsittaci in Genbank, a pair of primers were designed to detect the samples collected from 5pig farms where the chlamysis antibody was positive. The PCR positive samples were usedto isolate Chlamydia by chicken embryos. The Chlamydia isolates were identified bymicroscopical observation, inoculation in bacterium media, neutralization test,sulfamlamide sensitivity test and iodine stained-test. And ELD50s of the isolates were detected. The results showed that three samples were turned out to be PCR positive. Twostrains of Chlamydia (NN1, YL1) were isolated from the three samples and identifiedChlamydia psittaci. ELD50s of the two strains NN1 and YL1 for chicken embryos were10-13/0.4mL. |
