Concentrations of many kinds of mineral elements were detected in the serum of jugular vein and mammary vein of the cow suffered from alcohol positive milk(APM) in order to study the mechanism of APM further.Meanwhile,the anti-oxidation indexes were detected in the serum of jugular vein and mammary vein.The varation of trace elements and metabolism of free radicals were emphatically studied.The relation between mechanism of trace elements and anti-oxidation indexes was also discussed for probing into the relation between mammary gland metabolism and the whole metabolism.The results showed:1.Concentrations of Ca,Mg,Na,K were analyzed in the serum of jugular vein and mammary vein.The results showed:The concentrations of Ca,Mg,K in the serum of mammary vein were significantly lower than those in the serum of jugular vein in the normally secreted cow ( P<0.05 ) while the concentrations of Na were very significantly ( P<0.01 ) lower. The concentrations of Ca,Mg in the serum of mammary vein were very significantly lower than those in the serum of jugular vein in the cow suffered from APM(P<0.01),while the concentrations of Na,K were significantly(P<0.05)lower.Compared with the negative milk cow, the concentrations of Ca increased remarkably(P<0.05),concentration of Mg had no difference, the concentrations of Na decreased remarkably(P<0.05),while the concentrations of K decreased very remarkably(P<0.01)in the jugular vein of APM . Compared with the mammary vein of negative milk cow, the concentrations of Ca,Na increased but not significantly, the concentrations of Mg decreased significantly(P<0.05), the concentrations of K decreased very remarkably (P<0.01) in the mammary vein of APM.The result indicated that the excess Ca,Mg in the milk of APM broke the stability of casein ,the decrease of Na,K can not maintain the satbility of milk effectively.2.Concentrations of Zn,Cu,Mn,Fe were analyzed in the serum of jugular vein and mammary vein.The results showed:The concentrations of Zn in the serum of mammary vein were significantly lower than those in the serum of jugular vein in the normally secreted cow(P<0.05) while the concentrations of Cu,Mn,Fe were not different significantly. The concentrations of Zn in the serum of mammary vein were very significantly lower than those in the serum of jugular vein in the cow suffered from APM(P<0.01),while the concentrations of Cu,Mn were significantly(P<0.05)lower, the concentrations of Fe were not different significantly.Compared with the negative milk cow, the concentrations of Zn,Cu decreased remarkably(P<0.05),concentration of Mn had no remarkable difference, the concentrations of Fe increased remarkably(P<0.05)in the jugular vein of APM . Compared with the mammary vein of negative milk cow, the concentrations of Zn decreased very significantly(P<0.01), the concentrations of Cu,Zn decreased significantly(P<0.05), the concentrations of Fe increased very remarkably(P<0.01) in the mammary vein of APM.The result indicated that the balance of both mammary and whole body metabolism was in disorder,that was in the situation of oxidate.That needs the trace elements enter the reaction of maintaining free radicals metabolism balance which can enhance the activity of anti-oxidate enzyme,the trace elements which can enforce the oxidate of the free radical needed to be reduced.This may decrease or increase the relevent trace elements.3.The variety of anti-oxidate indexes such asGSH-Px,SOD,MDA,ROS,T-AOC were analyzed in the serum of jugular vein and mammary vein.The results showed:The concentrations of MDA and ROS in the serum of mammary vein were significantly higher than those in the serum of jugular vein in the normally secreted cow(P<0.05) while the activities of SOD,GSH-Px were lower significantly(P<0.05). Meanwhile,the activities of TAOC were appreciably lower but not different significantly(P>0.05).The concentrations of MDA and ROS in the serum of mammary vein were very significantly higher than those in the serum of jugular vein in the cow suffered from APM( P<0.01 ) ,while the activities of SOD,GSH-Px,T-AOC were very significantly(P<0.01)lower.Compared with the negative milk cow, the activities of SOD,GSH-Px decreased remarkably(P<0.05)in the serum of jugular vein in the cow suffered from APM while the concentrations of MDA and ROS were significantly higher(P<0.05), the activities of TAOC were appreciably lower but not different significantly(P>0.05). Compared with the mammary vein of negative milk cow, the activities of SOD decreased significantly(P<0.05), the activities of GSH-Px decreased very significantly(P<0.01)while the concentrations of MDA and ROS increased very remarkably(P<0.01) in the mammary vein of APM. T-AOC were very significantly(P<0.01)lower.The result showed:The anti-oxidate ability of mammary gland is not very good,but it can maintain the normal secretion by protective substitution.But mammary gland of the APM cow induced excess free radicals,lowered the total anti-oxidate ability so that the metabolism of free radicals is out of balance seriously.Consequently it could incur oxidized injury,which makes the secretion of the epithelial abnormal.
