Composition of low molecular weight glutenin subunits and gliadins were investigated with thirty common wheat cultivars which are widely grown in Shangdong province. In our study we used acid polyacrylamide gel electrophoresis (A-PAGE) and SDS-polyacrylamide gel electrophoresis (SDS-PAGE), analysed Sedimentation test, Protein content, Mixing time, Stability, Max-resistance to extension, Farinograph Quality Number and Extensogram area of the 30 common wheats. They have close relationship with the technology properties of noodle wheat cultivars. And we cloned a LMW-1 and a LMW-2 gene of common wheat by genomic PCR. The main results are as follows:1 LMW-GS and Glidian composition of 30 common wheatsAfter analysis of the results of SDS-PAGE and A-PAGE of 30 common wheats, we found that 17 cultivars possessed the component LMW-1/γ-42,they occupied 56.7% of the total. The other 13 possessed the composition of LMW-2 andγ-45, they occupied 43.3%.2 Relationship between LMW Glutenin Subunits/gliadins and gluten strength in common WheatProtein content, Mixing time, Stability, SDS-sedimentation test, Farinograph Quality Number, Max-resistance to extension and Extensogram area were analyzed. Results clearly showed that quality differences in common wheat were associated with the composition of LMW glutenin subunits and glidians. Cultivars possessing LMW-2 andγ-45 had better Processing Quality than the cultivars with LMW-1 andγ-42.3 Cloning of low-molecular-weight glutenin LMW-1,LMW-2 genes in common wheat and their relationship with quality properitiesWe used the specific primers of LMW-1 and LMW-2 in durum wheat and cloned a LMW-1gene and a LMW-2 gene of common wheat by genomic PCR. After sequencing the amplification product from Taishan201 is 882 bp (named PL1, GenBank accession Number DQ657349) and the product from PH82-2-2 is 888bp (named PL2, GenBank accession Number DQ657348).We compared the two amino acid sequence. The deduced mature amino acid sequence of the gene PL1 starts the sequence with METRCIPGL- , it is in the range of LMW-m, while PL2 starts the sequence with SHIPGL- directly, it is in the range of LMW-s. The deduced mature amino acid sequence of the gene PL1 has a deletion of 16 amino acids in the C-terminal domain.Furthermore, the deduced amino acid sequences of PL1 and PL2 in common wheat were compared with the sequences of LMW-1(GenBank number Y18159)and LMW-2(GenBank number AJ007746)in durum wheat . They have similar structure of the sequence. PL1 has two deletions in the repetitive domain and PL2 has three deletions in the repetitive domain compared with the genes of LMW-1 and LMW-2 in durum wheats. And the deduced amino acid sequence of PL2 has an insertion in the C-terminal domain, including 16 amino acids compared with the genes of LMW-2 in durum wheat. The other sequence has high identity.
