| Propolis is an natural immunological enhancement. Factors, which influences property of propolis extraction were studied in the experiment, such as different polar solvent, different concentration ethanol, different temperature. The virus strains in the research were isolated from Shandong chicken houses suffering borderline ND and EDS-76. Simultaneity, bivalent proplis inactivated vaccine was made successfully by Shen-zhiqiang know-how with a certain proportion of the antigen and ethanol extract of propolis. Conteracting toxic substances were carried in this article,we get minimum vaccination dose, immune duration, the storage period. So does effectiveness test. Humoral immunity and cellular immunity were further examined by HI and lymphocyte transformation.Its each experimentral result as follows:1. The content of effective component were different when propolis was extracted by solvent arrange from low to high according to their polarity.In addition, the CEC rose as the solvent poloarity rose. Furthermore, when EtOH was selected as solvent, concentration of EtOH mainly contributed to CEC. Processing temperature has some effects on CEC, but not as markedly as concentration.2. According to a series of tests to 5 batches vaccine, such as aseps;s,inactivation,animal security, the safety factor of vaccine reached 100%. At one time, the acute toxicity tests were studied on immune animals, and we got the minimum immunity dose of 0.3ml per chicken, the immunity protection persistent period of more than 6 months.In addition,the storage period of vaccine is 12 months in 4℃～8℃and 6 months in room temperature, then the immunity protection rate is 100% and more than 92% respectively.It showed that the antibody produced after the vaccine immuned was earlier, the titer was higher and the duration was longer than oil adjuvant vaccine. Probably the most important benefit one can get from using proplis vaccine in prevention of ND and EDS-76 is that it can improve the host cellular imunity leap and bounds, which may lead a central role in the preventive of ND and EDS-76 as can be reflected both in the level of lymphocyte proliferation as well as the the other two parameters got from Comparation of spleen. |
