The Study Of SRAP Molecular Markers In Sugarcane Mutant

SRAP (Sequence-related amplified polymorphism) molecular marker is a new molecular marker developed by Li and Qurios. It is simplicity, reliability, moderate throughput ratio and facile sequencing of selected. The study used this technique analysis donators and mutants of sugarcane , looked for SRAP marker between donators and mutants. The main results are as follows:1. Filtered 21 couples primer entities with polymorphism in 104 couples SRAP primer entities through SRAP amplification. Selected five couples primer entities with stable polymorphism between the mutants and their donors.2. Established the best SRAP amplification system of sugarcane. Searched out the best temperature of anneal, unlink and number of circle, reaction procedures.3. Searched out stable SRAP marker between donators and mutants. A 750bp SRAP marker was found in the mutant of the G11Bian. A 500bp SRAP marker was found in the mutant of the Luo Gu Zhe . A 750bp SRAP marker was found in the mutant of the Tiaowen F134.
