With the extending of rape's use, the planting areas become biggeryear after year. However, the rape diseases caused by bacteria, such asXanthomonas campestris pv. campestris (Xcc), are very severe. Thegeneral chemistry control methods should result in the environmentpollution. Harpin is a kind of protein produced by the specific bacteroidalphytopathogen in nature and encoded by hrp genes. Harpin can trigger acomplicated physical defense response against the epiphyte, bacterium, andvirus diseases, although harpin itself has no bactericidal ability. Therefore,it should be a good way to transfer the hrpW gene of Xcc into Brassicanapus L. In this study we carried out the experiments as follows:(1) The recombinant plasmid pBIhrpW was constructed by usingplant expression vector pBI121 with CAMV35S and NOS-term aspromoter and terminator respectively. The plasmid pBIhrpW wastransferred into Agrobacterium tumefaciens. The function of hrpW genewas identified through pressure infiltration assay on the non-host plantpepper lines (Capsicum annuum cv. ECW-10R) and the host plant radish(Raphanus sativus L. var. radiculus Pers.) seeding.(2) An efficient and stable regeneration system was established byusing cotyledonary petioles from 5~6 days' aseptical plantlets of B. napus as explants. The factors affecting transformation of cotyledonary petiolesof B. napus mediated by A. tumefaciens were explored.(3) The hrpW gene of Xcc was transferred into Brassica napus L.mediated by A. tumefaciens. Altogether 8 Kanamycin resistant plants wereregenerated. PCR detection showed that the hrpW gene had been integratedinto B. napus L. genome in 6 transgenic rape plants.Our results showed that it is possible to cultivate the new B. napus L.rape through the biological technology method. It provides theoryreference for new disease control strategy. Furthermore, the obtainedtransgenic plants provide a good vessel to investigate the pathogenesis ofXcc.
