Cotton is a very important economical crop for its role of producing crude fibre.the sector of weave introduced an effective and rapid spinning technology,it will raise much more requirement to the attribute of its fibre.behaving the research of gene function in cotton will throw light to genetical improvement with cotton obviously.Gibberellic acid is a large family which comprise many terpene compounds,it plays an important role in regulation of complete life cycle ,such as in ,GA binds to the acceptor on cell membrane,and then transfer its signal to downstream through a series of signal molecules for regulating the growth and development of plant.But GAI is a negative regulated factor to the pathway of GA signal transduction.The known result indicated the mechanism underlied in the development of cotton fibre is similar to the development of burr in Arabidopsis thaliana.therefore,To research the influence of GAI gene and GAIL(which showed DELLA protein lacked) on the development of burr in Arabidopsis thaliana will provide theoretical basis on dissecting the mechanism underlied in the development of cotton fibre.For the purpose of identifying the funtion of GAI gene and GAIL in cotton,our research transfer GAI gene and GAIL of Gossypium hirsutum L and empty carrier PBI121 into Arabidopsis thaliana by the method of vacuum filtering. As a result, six lines of transfer GAI, eight lines of transfer GAIL and five lines of transfer empty carrier PBI121 have been filtrated in T1 generation of transgenic plants . T2 generation transgenic plants phenotype observed found : There are noticeable changes in seed germination , root development level , sprouting and flowering time and height, crown diameter and the number of branches; However no obvious impact on the development of burr; and that in the three genotypes plant seedlings some lines have already emerged albino or reddish-brown, Transplanting can not survive. Analyzed gene expression by effectively prevent the plant genomic DNA contamination TR-PCR new technic ,useing homemade electric grinding and improved plant tissue polishing methods indicates GAI gene and GAIL gene have been transcripted in T2 generation. therefore infer GAI gene and its protein deletions GAIL may correlated with the seed germination , root development level , sprouting and flowering time, height, and the number of branches of cotton . In addition, abnormal phenotype happened in homozygous of GAIL.This study not only establish a base for further study cotton GAI gene and its protein deletions GAIL gene foundation, but also provide a way of grinding a small quantity of plant tissue and an effectively prevent the plant genomic DNA contamination TR-PCR new technic, theoretical and applied with a certain value.
