Expressing homogeneous extraneous source calcium metaboliable regulators gene in vivo can make up corresponding material synthesis and the secretion insufficiency in layers. Artificial short-term regulating layer calcium nutrition metaboliable can provide the foundation for the gene enhancement therapy in nutrition metabolic disease. Firstly, the chicken calcium metaboliable regulators eukaryon expression plasmids, including pCEP4-CT, pCEP4-PTH and pCEP4-CaBP-D28k are tested by enzyme digesting and the appraisal prepared. Then, two experiments are carried out to consider the effects of chicken calcium metaboliable regulators eukaryon expression plasmids under two different scales of experimental animals. The determinations of CT and PTH in serum, egg production performance, egg shell quality, bone mass contents, metaboliable experiments and body-weight are carried out.1.In Bench Scale Test, CT in serum of CaBP Group at 2nd wk is significantly higher than that of Vector Control Group (P<0.05); PTH Group at 3rd wk is extremely higher than that of Blank Control Group (P<0.01), significantly higher than that of Vector Control Group (P<0.05), CaBP Group is significantly higher than that of Blank Control Group (P<0.05); Treatment groups at 4th wk are extremely higher than that of Blank Control Group (P<0.01), CT Group is significantly higher than that of Vector Control Group (P<0.05). PTH in serum of CaBP Group at 2nd wk is extremely higher than that of Blank Control Group (P<0.01); CT Group and CaBP Group at 6th wk are significantly higher than that of Blank Control Group.2.In Bench Scale Test, egg production rate of CT Group at 3rd wk is significantly higher than that of Blank Control Group (P<0.05), extremely higher than that of Vector Control Group (P<0.01); CT Group and CaBP Group at 4th wk are significantly higher than that of Vector Control Group (P<0.05). Egg-weight and egg shape index of groups between various weeks are not significantly different (P>0.05). In Pilot Scale Test, egg production rate of CT Group at 2nd wk is significantly higher than that of Blank Control Group (P<0.05); CT Group at 3rd wk is extremely higher than that of Blank Control Group (P<0.01). Egg-weight of treantment groups compared with control groups are not significantly different (P>0.05); Egg shape index of PTH Group and CaBP Group are significantly higher than that of Blank Control Group (P<0.05), treantment groups compared with oriental wk are not significantly different (P>0.05).3.In Bench Scale Test, eggshell intensity of groups between various weeks are not significantly difference (P>0.05). Relative eggshell-weight of groups between various weeks present large shaking trend generally. Eggshell thickness of treantment groups between various weeks present upwards shaking trend generally. In Pilot Scale Test, eggshell intensity of CaBP Group is higher than that of control groups (P>0.05). Relative eggshell-weight of PTH Group and CaBP Group are extremely higher than that of Blank Control Group (P<0.01), CT Group is significantly higher than that of Blank Control Group (P<0.05), treantment groups are extremely higher than that of oriental wk (P<0.01). Eggshell thickness of CaBP Group is significantly higher than that of control groups (P<0.05).4.In Bench Scale Test, skeleton targets of groups between various weeks are not significantly different (P>0.05). In Pilot Scale Test, BMD of PTH Group is significantly higher than that of CaBP Group (P<0.05); Ash of groups are not significantly different (P>0.05); Ca of CaBP Group is significantly higher than that of Blank Control Group (P<0.05); P of groups are not significantly different (P>0.05). 5.In Bench Scale Test, GE metaboliable rate of CT Group and CaBP Group are significantly higher than that of Blank Control Group (P<0.05); treatment groups are extremely higher than that of Vector Control Group (P<0.01). CP metaboliable rate of CT Group is significantly higher than that of Blank Control Group (P<0.05). Ash metaboliable rate of CT Group and CaBP Group are extremely higher than that of control groups (P<0.01); PTH Group is significantly higher than that of Blank Control Group (P<0.05), extremely higher than that of Vector Control Group (P<0.01). Ca metaboliable rate of CaBP Group is significantly higher than that of Vector Control Group (P<0.05). P metaboliable rate of CT Group and PTH Group are significantly higher than that of Blank Control Group (P<0.05), extremely higher than that of Vector Control Group (P<0.01); CaBP Group is significantly higher than that of Vector Control Group (P<0.05).6.In Bench Scale Test, body-weight of weeks between various groups are not significantly different (P>0.05). In Pilot Scale Test, body-weight of CaBP Group is extremely higher than that of Blank Control Group (P<0.01), significantly higher than that of oriental wk (P<0.05); CT Group is extremely higher than that of Blank Control Group (P<0.01), PTH Group is significantly higher than that of Blank Control Group (P<0.05).After the injection of plasmids, obviously changes of CT and PTH in serum show that the plasmids have already expressed successfully by blood targets. By egg targets, all three kinds of plasmids can slow down effectively or counter the depression of egg production rate and eggshell quality. By skeleton targets, all three kinds of plasmids can enhance the bone mass contents significantly. By metaboliable targets, all three kinds of plasmids can enhance the efficiency of nutrients in feedlots, increase the body mass, and slow down or reversal the body-weight lose.
