Paralichthys olivaceus is one of the most valuable fish in Hebei province.At present , this kind of fish is breeded in a large scale in coastal aear of north China.Along with the development of the indoor breeding and the pollution of the sea, amount of pathogenic bacteria which disserved Paralichthys olivaceus has appeared.All of it caused a serious expense of the economic.The experinment isocilated the pathogenic bacteria from diseased Paralichthys olivaceus .We used kinds of biochemical methods including Gram staining,the detection of its motility,medicine sencitivity and so on;We also detected these bacterias by using polymerase chain reaction based on the aer gene and the 16s rDNA sequence of Aeromonas hydrophila reported on NCBI.The result of the molecular detection was identified with the biochemical detection.We used Aeromonas hydrophila which we separated from the diseased Paralichthys olivaceus to made a inactivated whole cell bacterin .We injected this vaccine in the mice and the Paralichthys olivaceus .Both the mice and the Paralichthys olivaceus gained a good protection of 80% and 70%.The fish with dipping immersion gained a good protection of 85% ,and the incidence of disease was 6%.We detected the antibody level in the antiserum by agglutination reaction and indirect ELISA which seemed to be immunogenic crucially.We anatomized the mice then we found that all the purtenance of the immunzied mice are healty.All of this proved that the vaccine is effective.
