Genetic variance and genetic identity of alfalfa in morphological level and DNA level are studied by morphological and RAPD techniques using 9 varieties (germplasm).Abundant genetic variance is found in alfalfa through research on the morphologic traits.The trait with the largest variance is leaf area, and the trait with the larger variance is leaf width in 9 Alfalfa Varieties(germplasm) Correlation-analysis indicated that all the variance levels have significant correlation among the traits except node number and average internode length. Main component analysis indicated that the stem length, node number, average intemode length, leaf width factor and shape factor made up the first component and they play a important roles in the total morphological variances.Genetic diversity of 9 varieties (germplasm) of alfalfa is analyzed using RAPD technique. The result showed that 96 loci and 64.22 polymorphism loci has been amplified using 10 primers. Percentage of polymorphic loci is 66.90%. The diversity of Nei index evaluation is lower than Shannon index evaluation. But the differentiation by Nei index was close to differentiation by Shannon index. M. falcata No.2 and M. falcata No.3 exhibit high level of genetic diversity. Cluster analyses based on RAPD markers and based on morphological level show that the most genetic distance is between M. Purple flower and M. Varigated flower.Correlation analysis between morphological traits index and genetic diversity index indicated that morphological traits index and genetic diversity index have not correlation.
