Drought stress often affects growth of wheat and causes the grave reduction of output of crop. Crop's growth and output are outcome of inherits gene expressing in specially appointed environment, that whose actual output and latent productive capacity differ by the damage of bad environment and drought is one of very important factor. Carrying out studies on crop drought resisting has very important significance to relieving water resource deficiency, improving WUE of crop and realizing water safety and food security. Hereditary characters of wheat on drought resistance and water saving is very complicated. From breeding tactics, breeding means and breeding technology, improving reliability of choice and authenticate index of hereditary character can relieve crop water saving effect and it is the important breakthrough point in drought resistance and water saving breeding theory and breeding practice.The endurance ability of the wheat under the water stress condition originates from whose inherent molecule basis. So this ability of wheat differs by different cultivars. Under same arid condition, drought-resistant cultivar may produce more response protein to fight against the threat of lacking water. Response protein induced by drought plays important protection role and can improve plant the endurance ability to drought stress. In recent years, our country research in the respect of coercing factor to inductive protein has got very big progressing. Drought induces plant to produce peculiarity protein, which is regarded highly gradually. Response protein induced by drought already becomes a machine-made hot spot fighting drought to crop in studying adapting to arid adverse circumstances gene expression.Thirty winter wheat cultivars were selected in the research. The pertinence between water stress induced response protein and drought resistance and the expression of D-response protein during seedling stage are studied. By drought resistance identification of wheat, dealing the seeds with water or PEG-1.0MPaPEG6000 and SDS-PAGE method,research the D-response protein of different wheat cultivars which the seeds were respectively deal with water and PEG, ascertain the relationship between the D-response protein and drought resistance. The result indicated that after being water stress, all the best and the most of better drought-resistance species appear the D-response protein stripe whose molecular weight was appropriately 66.2 KD, but all sensitive species did not appear this stripe in the water stress and no stress condition. That explained that the D-response protein induced by water stress during winter wheat seedling stage has close relation with the drought resistance of cultivate.In order to study on the gene of D-response protein, the male parent which has D-response protein, the female parent which has no D-response protein and their F2 segregated population are chosen as experiment materials, that being based on the result of experiment during the earlier stage. Result of SDS-PAGE test to the protein of the single plant of F2 population indicates that the D-response protein of wheat is controlled by one-allele gene. Adopting SSR technology union BSA method, DNA of D-response protein pool and no D-response protein pool are established by using D-response protein and no D-response protein plants of F2 population. Among the 31 pairs of micro-satellite primers, one marker is found polymorphism between the two pools. Linkage analysis showed that micro-satellite marker Xgwm156 is linked with the gene of D-response protein, as showed the micro-satellite marker Xgwm156 is a molecular target of drought resistance identification and chosen.
