Content: Cryopreservation has long been considered an important tool for the long-term storage of plant germplasm. This paper outlines new developments in the last 10 years of plant.Pavlova viridis and Isochrysis galbana 3011 were cryopreserved in liquid nitrogen using encapsulation-vitrification.Algal cells in early stationary phase were encapsulated in 3% Ca-alginate beads with 30‰NaCl,3 million cells in one bead.Beads were loding with 60% PVS ( PVS Isochrysis galbana 3011 :30%(w/v)glycerol, 20%(w/v)ethylene glycol and 10%(w/v)dimethyl sulfoxide in f/2 medium; PVS Pavlova viridis : 30%(w/v)glycerol, 20%(w/v)ethylene glycol,10%(w/v)dimethyl sulfoxide and polyvinylpyrrolidone in f/2 medium)at 20℃, then dehydration with 100% PVS at 0℃prior to a direct plunge into liquid nitrogen and warm in 40℃water bath. The main factors influencing the cell viability, such as the composition and the concentration of loading solution and loading time, the time of dehydration, the concentration of washing solution and washing time were studied. The results are as follows:1. Isochrysis galbana 3011: The result showed that the high viability of 45% was obtained when the beads containing the algal cells were loaded for 60min at 20℃, then dehydrated for 60 min at 0℃prior to a direct plunge into liquid nitrogen and washed by 0.5mol/L sorbitol for 60min after warming in 40℃water bath.2. Pavlova viridis: The result showed that the high viability of 40% was obtained when the beads containing the algal cells were loaded for 180min at 20℃, then dehydrated for 60 min at 0℃prior to a direct plunge into liquid nitrogen and washed by 1.0mol/L sorbitol for 40min after warming in 40℃water bath.The method of encapsulation–vitrification do not need special equipments, simplifies the operation procedure of cryopreservation, so it is a potential method for preservation of I. zhanjiangensis and other algae.
