In veterinary clinics,canine breast tumor was a common tumor disease in femaledogs. It shared many similarities with the breast tumor in humans, such as theetiology, the characteristics of tumor tissue, the therapeutic in medicine, themammectomy and the prognosis. Therefore, canine of spontaneous breast tumor hadbecome the most excellent models for human studying breast tumor disease. Thepathohistology method, the immunohistochemistry method and the cytobiologytechniques were employed in the experiment to carry out the investigations on thespontaneous canine breast tumor and the spontaneous cell lines, in order to provide thediagnosises and the treatment basises to canine breast tumor.It was adopted in the experiment that four cell lines of CHMp,CHMm,CIPp andCIPm which were isolated and cultured from the primary lesions and the metastaticlesions in the spontaneous cases with breast tumors. The research objects were selectedfrom the cell lines with above 120 generations and steady growth traits. The cells werecultured with the digestion method, and the results indicated that the growth curveaccorded with "S" shape,and the doubling time respectively is21.3h,35.3h,42.3h,49.1h. The cell's morpHous was not significantly deviated after highpassages. Using the MTT examination method and the soft agar-agar culturecomparative analysis indicated that CHMp,CHMm were better than CIPp, CIPm in thecell vigor and in the increasing ability. The results indicated that there were significantdifferences on the cell biological characters in different cell line sources. 10 cases ofsurgical operations excised doubtful canine breast tumor specimens were classified bythe pathohistological observation. The immunohistochemistry method was used in theexperiment which used two different kinds of rabbit anti-mouse polyclonal antibody todetect specimens and compare antibody expressions and pathology types. The resultsindicated that: malignant breast tumor was to 50%, in which mammary infitratingductal carcinoma was to 30%; benign tumor was to30%, in which non-breast tumor20%. The results showed that the rabbit anti-mouse polyclonal antibody p53 could react with the canine breast tumor tissues and the expression in breast cancer wassignificantly higher than benign breast tumor. CD44 expressed in the breast cancer, thebenign breast tumors and the non-breast tumor tissues,and it was not statisticlysignificant, p53 could be used as one of the indications for the animal prognosis.Thecellular immunochemistry analysis indicated that: p53, CD44 and VEGF had positivereactions with the different clinical significances: VEGF was not significantly differentin 4 kinds of cell lines which indicated that the functions and the actions mechanismwere similar in the matastatic group and in the primary group breast tumor cell; Theexpressions of CD44 had the differences in 4 kinds of cell lines, which indicated that itcould not be taken as the independent prognosis index to show the infusion andtransfusion which needed to the participation and regulation of other factors. Theweakest expression of p53 was in CIPm cell which had the weakest ability inactivation and increment, which was corresponded with the expression results intissues, and it was proved that the reliability of p53 taken as the animal prognosismarker.
