Bacterial wilt is a kind of ruinous soil-borne disease. As one of mostimportant plant diseases in distribution and losses in the world bacterialwilt still not been effectively controlled by pesticides and othermeasures.On this study specific primers (AU759/AU760) were designed to detect R.solanacearum by PCR. The result indicated, not only should to amplify the specificityfragment (281 bp) to R. solanacearum isolated from chilli pepper, also may R.solanacearum which from tomato, eggplant, potato, fructus momordica, tobacco andso on in many kinds of crops isolated. Our results offer a fast specific detectionmethod of R.solanacearum in various crops. The amplified 281 bp fragment wasfurther sequenced which is a part of the lpxC according to the blast search inGenebank. The IpxC gene encodes the UDP-3-O-Acyl-GlcNAc diacetylase enzymewhich catalyze the second step of the lipoid A, a essential substance of R.solanacearum, biosynthesis.Pathogenicity of 20 R. solanacearum strains were tested in tomato line L390, and 8virulence strains were achieved. Furthermore, RS-4 strain was selected for resistancescreening with 13 pepper varieties, which are the representative lines from AVRDC(The World Vegetable Center) genbank. Six pepper varieties with high resistance to R.solanacearum were obtained, which could used in future resistance breeding.
