Potato (Solanum tuberosum L.) belongs to Solanacase family. As less tsugiyo timeof potato culture in vitro and there are some difficulties in its cryopreservation, there-fore, the development of potato germplasm conservation is limited. This experimentwas studied on potato conservation time, changes of physiological and geneticstability under 4℃by plantlets. The aim is to prolong the conservation time of potatoplantlets in vitro and set up a suspension system for potato germplasm conservation,and afford theoretical basis and new method for decreasing it costs. The main resear-ch results are follows:1. Different concentration levels of CCC, PP333, MH and SUC have been added inMS culture medium with 4℃treaded. The resulted shows that tsugiyo time of potatoplantlet has been prolonged from 100 days to about 240 days as MS+CCC600mg.L-1with 4℃treaded. Moreover, the potato plantlets can be renewal growth quickly.2. physiological Changes of potato plantlets Conservation at low temperature werestudied. The results showed that low temperature can inhibit the growth of potatoplantlet obviously, the contents of chlorophyll, Vc, Pro and SOD in plantlets weredecreased during the preservation, but under 4℃were higher obviously; The activityof POD were increase in 60 days, then decreased quickly, but under 4℃were lower;The content of MDA was increase, but under 4℃were lower than 25℃.3. RAPD was first used to study genetic stability of potato plantlets which havebeen treated with 4℃and MS+CCC 600mg.L-1. The reaction system of potato DNAhas been optimized. As 120 bar random primer detection, there were no changes andvariation DNA fragmentation for potato DNA, the potato genetic character wasrelative stability.4. Minituber and elite seed field production experiment of potato were used todetection genetic stability of regeneration potato plantlet. Its leaf shape, plant shape,height, leaf length, yield of individual and many biological character have not founddistinct variation, the genetic stability and anticipate character were stability.
