In this study, in order to effectively transcribe dsRNA in plants, an inducer ofRNA silencing, CMV (Cucumber mosaic virus) partial movement protein gene(â–³MP) were in inverted repeat manner, the recombinant was then inserted intobinary vector pBIN438 under control of the 35S promoter. 34r transgenic tobaccoplants expressing CMVâ–³MP gene were generated by Agrobacteriumtumefaciens-mediated transformation, resistance and possible mechanism forresistance of these transgenic plants were evaluated.pBIN-CMV MP(i/r) was transformed into A. tumefaciens EHA 105 by tri-parentalmating method. The positive clones confirmed by PCR and digestion were used fortransformation of tobacco plants. The vector pBIN-CMV MP (i/r) was transformedinto tobacco by A. tumefaciens-mediated method via leaf disc, 30 kanamycin-resistanttransgenic plants expressing TMV MP (i/r) were obtained. PCR and Southern blotanalysis of transgenic plants showed that CMV MP (i/r) had been integrated intotobacco genome. 30 transgenic plants expressing CMV MP (i/r) were inoculated withCMV, symptom observation and ELISA detection showed that transgenic plants haveresistant to CMVOur results lead a way for plant virus control using dsRNA through PTGSapproach.
