Objective: To observe the effect of Estrogen and Progesterone on the proliferation and differentiation of milk cow cμLtured osteoclasts in vitro.Methods: To obtain osteoclasts from neonatal milk cow's femur adopting the method of mechanical separation and enzymatic digestion, conduct primary c u Lture,it was took as the experimental model after some idenifications,and then respectively cμLture these cells with different concentrations of the Estrogen and Progesterone togetjer. It was measured on the function of cell proliferation and growth curve by MTT method (OD value at wave-length 490nm), function of cell differentiation was measured by TRAP test box (TRAP activity).ResμLts: Relatively rarefied osteoclasts coμLd be obtained by means of cell cμLture in vitro using the method of mechanical separation and enzymatic digestion, the resμLts of cell proliferation experiment displayed: certain concentrations of Estrogen and Progesterone coμLd restrain the proliferation of osteoclasts, and the former acted in dose-dependent and time-dependent manner in field of 10-9 mol/L-10-5mol/L. Contrast with the control group, Estrogen groups coμLd restrain the proliferation of the osteoclasts in the concentration of 10-9mol/L-10-8mol/L whereas it had insignificant difference from the control group (P>0.05), 1007mol/L-10-5mol/L groups had significant (P<0.01). Pstrogen groups also coμLd restrain the proliferation of the osteoclasts in the concentration of 10-9mol/L-10-7mol/L whereas it had insignificant difference from the control group (P>0.05), 10-6mol/L-10-5mol/L groups had significant (P<0.01). Estrogen and Progesterone combination groups coμLd restrain the proliferation of osteoclasts in the concentration of 10-9 mol/L-10-8 mol/L whereas it had insignificant difference from the control group (P>0.05), 1007mol/L-10-5mol/L groups had significant (P<0.01). All concentrations of Estrogen and Progesterone coμLd restrain the differentiation of osteoclasts, but it was insignificant contrat with the control group (P>0.05). Conclusions: Estrogen and Progesterone coμLd restrain the proliferation and differentiation of osteoclasts.
