The thesis includes two parts: the copy number detection of KIT gene in Wuzhishan pigby quantitative real-time PCR; association analysis between porcine PSME1 gene andimmunity traits.Part one: Wwzhishan pig, well-known, Chinese-special local breed, ideal medicalexperimental animal modle, now be used for studies of pharmacology, pasturage, veterinary,plays an important role in heterogenous organs transplantation. Wuzhishan pig is originatedfrom Hainan province, south China. Since 80s last century, the pigs was introduced intoInsitute of Animal Ssiences, Chinese Academy of Agricural Sciences and begun to long-terminbreeding. The main coat color of Wuzhishanpig is black, belly and insides of four legs arewhite, mane is black and brown. The special coat color is one of characteristic traits in thepigs which can be used to breed determination. The dominant white coat color in pigs iscontrolled by the dominant white (I/KIT) locus. Three alleles have been documented at thelocus: Dominant white (I), Patch (I~p), and the recessive wild type allele (i). Different alleleshave different number of KIT gene copies. Both alelle I and I~P carry two KIT copies,therefore alelle i carry one KIT copy. The KIT gene codes mast/stem growth factorreceptor. Normal expression of KIT and its ligand MGF is essential for migration andsurvival of neural crest-derived melanocyte precursors. When the KIT gene was expressedin normal, pigs showed black coat color. Variations in molecular sequence or copynumber of KIT gene cause a loss or a marked reduction in the number or melanocyteprecursors in the embryo and finally lead to loss of the coat color. Here copy numberdetection of of KIT gene was performed using Quantitative real-time PCR to providesome evidence for genetic mechanism of Wuzhishan pig coat color formation. Results:one KIT copy was found in Wuzhishanpig, the molecular orgnization of the gene inWuzhishanpig was evidently similar to its in Duroc pig(one KIT copy).Part two: PSME1 gene encodes PA28 a submit. PA28 protein is activator ofimmunoproteasome, which is responsible for the generation of cytosolic peptides present byclassâ… molecules. To study the relationship between PSME1 gene and pig breeding fordisease resistance, SNP detection in porcine PSME1 gene was performed using DHPLC, pigpopulation was genotyped by PCR-RFLP, and association analysis with immune traits was also done by SAS6.0. Results: Tatol 6 SNPs was found in porcine PSME1 gene; a SNP in3rd intron was associated with RBC, HCT and IgG.
