Classical swine fever (CSF) is a highly contagious disease in pigs caused by the CSF virus. It wasfirst discovered in the state of Ohio of America. The disease spread worldwide and its main characteristicsof are high morbidity and mortality. Bring a severe economic losses to the pig farming worldwide. CSFspread severely in our country and about 39.65% of pigs died of disease died of CSF. The economic lossesreach a hundred million RMB. The Hog cholera lapinized vires vaccine was developed in 1956 in ourcountry and was accepted the most safe CSF vaccine using all over the world. Many countries haveeliminated the CSF by this vaccine. Although our country also get a big achievement in prevention of CSFusing this vaccine but the CSF are still prevailing in most field of our country. We did not control theprevail of CSF by injecting vaccine in a large area. This lead us to doubt the immuno-efficiency of CSFlapinizated low virulent vaccine.Multitudes of domestic and abroad researchers made a lot of study on the humoral effect of CSFvaccine and hold the same result that the pigs immunized by Hog cholera lapinized virus vaccine canproduce neutralizing antibodies and provide a complete protection for CSF. But few research about theeffect of cell mediated immunity by advanced immune. Much abroad studies were to illuminate theimmunity mechanism by strong virulent vires and domestic studies were mock-up test by cony pigs or micethat different from the pigs themselves. Let along the effect of cell-mediated response in pigs after vaccineimmunization and whether the comparison of difference between advanced and normal immunization isnotable or not.In this study, we develop a track detection for ultra pre-immunized piglets using the traditionalmethods of white blood counting,differential count,lymphocytes proliferation test and Flow cytometrytechnique to observe the number of white blood cells and lymphocytes, the mitogenic responsibility oflymphocytes to PHA and the level of CD4~+,CD8~+ lymphocytes subsets and IFN-γrespectively. Then take acompared analysis with the normal immunized group and control group and give an objective evaluation tothe immnunal effect of CSF vaccine from cellular and molecular level.Study results demonstrated that the number of peripheral blood white blood cell, lymphocytes, CD4~+and CD8~+ lymphocytes increase obviously from pigs vaccinated at 0 day and 60 day with CSF vaccine. ThePBMC also respond to PHA obviously. The level of IFN-γalso increase obviously especially after thesecond immunization at 60 day. There is a significant difference between items in test group and controlgroup (P<0.05). But there is no significant difference between the ultra pre-immune group and normalimmnune group at the total immune level (P>0.05). The results shown that CSF ultra pre-immunity can induce and early cellular immunity and construct an defensive barrier to the CSFV.
