Porcine growth hormone (pGH), consisting of 191 amino acids, secreted by the anterior pituitary intothe circulation, binds membrane receptors in target tissues to stimulate body growth. Lack of GH has shortstature and GH therapy given before puberty increases their rate of growth and ultimate height. Studies onanimal further support an important role of GH in the promotion of growth. In addition, GH has importantmetabolic actions. In human and animal, exogenous GH decreases fat and increases lean body mass. pGHhas a function of promoting protein anabolism through accelerating amino entering cells and RNAsynthesis, pGH not only accelerates bone growing, but also affects development of many tissues. As resultof accelerating fat's catabolism, pGH increases muscles rate and feed efficiency.PGH was isolated initially from the anterior lobe in pituitary. Because of the great economic value,pGH has been industrially produced by genetic engineering in 1983, which refer to some technology, suchas gene regulation, protein expression, protein separation and protein purification, pGH is very low inproduction, biological activity, and pGH need to be injected by continuous intramuscular injection withlarge dose or subcutaneouly embedding. These limit application of pGH. With development of hormoneimmunology, the considerable experiments have been conducted to change the balance of internal secretionfor producing the significant effect of physiology.The objective of this study was to prepare anti-idiotype antibody of porcine growth hormone (pGH)from the goat immunized with monoclonal antibody against pGH(anti-pGH McAb), to determinelocalization of its intern image in porcine liver cells by immunefluorescence technology and to evaluate itsactivity of promoting growth in mouse. Anti-pGH McAb labelled with FITC was bound to growth hormonereceptor (GHR) of porcine liver cells, and revealed with confocal laser scanning microscope (LCSM). Theimpact of pGH anti-idiotypic antibody on growth was tested in mouse. The results indicate that pGHanti-idiotypic antibody mimics the function epitope of pGH and has an analogous structure of pGH, whichcan bind the receptor of GH in porcine liver cells, pGH anti-idiotype antibody displays the activity ofpromoting growth in mouse.These results established the foundation of theory and technology for further developing the new-typesomatotropinic surrogate.
