| The materials of this experiment are p5CS-bar gene A.cristatum×A.desertorum cv.'Hycrest-Mengnong'and CBF4-bar gene A.cristatum×A.desertorum cv.'Hycrest-Mengnong'which bombardment changes by gene-gun. Fist of all, I carried out PCR detecting to identify and gain transgenic wheatgrass. Than, on the basis of this, I observed biology characters and determined drought resistance,salt endurance to gain good transgenic wheatgrass in order to provide the academic basis for further study. The results are as follows:1,The transgenic wheatgrasse plants were carried through molecular detecting. The results showed that,①In 95 p5CS-bar gene transgenic wheatgrasse plants, the number of transform bar gene plants are 26 and the frequency is 27.37%, the number of transform p5CS gene plants are 49 and the frequency is 51.58%, the number of co-transformation are 15 and the frequency is 15.78%;②In 378 CBF4-bar gene transgenic wheatgrasse plants, the number of transform bar gene plants are 29 and the frequency is 7.67%, the number of transform CBF4 gene plants are 31 and the frequency is 8.20%, the number of co-transformation are 5 and the frequency is 1.32%.2,The biological characters of p5CS-bar gene transgenic wheatgrasse and control plants were observed and analysed that none but caudex have marked differences(0.05); The values of plant height,spike length,spike width and stem/leaf are less than control plants, leaf length and leaf width are more than control plants; Under the same condition of growth and climate to compare transgenic wheatgrass with control plants we can see that trning-green sage is advance, defer procreate period, long viridity period, growth speed become slow and the figure of curves is"S", more tillers.3,p5CS-bar gene transgenic wheatgrasse plants and CBF4-bar gene transgenic wheatgrasse plants were intimidated by drought and salt. From 6 aspects that are moisture status, photosynthesis, membrane oxidation, filter adjust, protective enzymes system and protein content to investigate. The results showed that,①the drought resistance and salt endurance of p5CS-bar gene transgenic plants are obviously higher than control, it is say that p5CS-bar gene have shift to wheatgrasse plants successfully and it has expressed in it.②the drought resistance of CBF4-bar gene transgenic plants are obviously higher than control, it is say that CBF4-bar gene have shift to wheatgrass plants successfully and it has expressed in it. |
