In this paper we studied tissue culture of Amorpha fruticosa and Tetraploid Black Locust, and established a good tissue culture system, optimized the rooting medium and resolved the problem of low rooting rate in vitro and low survival rate of transplant .The research project is meant to pick out the ideal species by applying 60Co-γrays to multiple bud clumps and seeds of Amorpha fruticosa in order to induce mutation. The results were as follows:Firstly, in the cource of tissue culture, the optimum differentiation culture medium substrate was MS+BA2.0-4.0mg/L for Amorpha fruticosa and MS+BA0.5-1.5mg/L for Tetraploid Black Locust. the optimum rootage culture medium substrate was MS+IBA0.2mg/L + NAA0.1mg/L + AC2g/L for two species.Secondly, Comparison of the two rapid propagation capacity, found both in the Bud induction of cytokinin sensitivity differences embodies their own genetic specificity. Meanwhile adventitious buds in high yield and the growth hormone sensitive areas shows that the similarity between the Cousin Plants.Thirdly, in the research of 60Co-γrays treatment on multiple bud clumps, the gamma ray restrain the growth, the higher the dose of gamma ray used, the wider the restrain effects of plants was. The optimum dose of 60Co-γrays treatment on the germination of Amorpha fruticosa was 20Gy.Multiple bud clumps were irradiated by 20Gy of 60Co-γrays, then cultured on media supplemented with varied NaCl concentrations continuously for 3 generations. Then, plantlets, which could actively proliferated on media containing 1.5% NaCl were obtained.fourthly,in the research of 60Co-γrays treatment on dry seeds,The germination rate, the plant height, and the rate of survival plant were studied. The results showed that: In experimental dosages, they showed paraboloid shape and had peak value with the increase of gamma-ray doses.Then,we identified that 400~500Gy is the semi-lethal dose. Through the cytology examine, we found aberrance chromosome such as chromosome bridge, fragement and lag chromosome and crokernel. We studied on the effects of radiation dosages of 60Co-γrays on three Physiological indexes such as the content of sucrose, the content of protein,and the content of chorophyⅡ. The results showed that there was not prominently negative correlation between three Physiological indexes and dosages of 60Co-γrays.
