5 melon parent lines included TopMark and H16 as male parent, 3-2-2, Tianshuai and Yucuias female parents and their hybrids were analyzed by RAPD and SSR markers, from clusteranalysis to find the reason of the difference inheritance between parent lines and hybrids. Withthe combination of morphologic performance, we want to discuss the inheritance relationshipbetween parent lines and hybrids.Estimated morphological characters in three different locations separately, the result showed:although difference of morphological characters among three locations existed, they had similartendency. Hybrids inherited more heredity from female parent than male parents, such asprematurity, fertility. It was similar with the results of molecular markers: F1 progeny and femaleparents had the most related relationship.After compare of parent lines and hybrids on morphological characters, Line Yucui had themost branch number and fruit number per plant; on the contrary H16 had the least. Progeny fromline Yucui and line TopMark had the most fruit weight per plant and the days to flower.Analysis of variation on fruit weight per plant, fruit number per plant and fruit weight permelon were prominence. Analysis of variation on morphological characters between energysaving greenhouse and plastic greenhouse were prominence. General Combing Ability (GCA) ofline Yucui and line TopMark had positive effects leading the progeny from Yucui and TopMarkhad the best performance on Special Combing Ability (GCA).Branch number and fruit weight per melon performed positive trendy in all hybrids, andprogeny from Yucui and TopMark had better heterosis on all morphological characters within allestimated hybrids.Different melons were amplified by 27 RAPD primers, total 44 alleles, which 19 of themhad polymorphism, and it was about 43.18%, revealed the average of 5.26 alleles per marker and3-8 bands per primer. The result of primer C01 has 3 amplified bands between 600-920bp, themost amplified bands were the result of primer AX-16, total 8 bands from 700bp to 2000bpwhich 5.26 bands per primer. After cluster analysis, the genetic distance between parent lineswere 0.09-0.41.4 hybrids have close genetic inheritance to female parent and 2 hybrids to maleparent.Using SSR markers to amplification, 33 alleles had polymorphism in total 39 bans analyzed by 14 primers about 84.61% which from 100-900bp. The result of primer N29 only has 1amplified band, the most amplified bands were the result of primer CMGA172, and total 7 bandsfrom 125bp to 180bp which 2.79 bands per primer. After cluster analysis, the genetic distancebetween parent lines were 0.14-0.73. Although there was some difference between RAPD andSSR markers analysis, the cluster analysis was similar. 3 hybrids have close genetic inheritanceto female parent and 2 hybrids to male parent.In a world, using molecular markers and combing ability analysis have a better result onanalysis of comparisons on inheritance between parent lines and progeny F1 on melon, giving abetter way to study inheritance breeding and parent selection on melon.
