This paper used quantitive genetics molecular genetics Biochemistrygenetic marker for studying heat-tolerance of Holstein. Material work andconclusion as follows:(1) Study on Relevant to Parameters Of Heat-Tolerance in Chinese Holsteinin Southern ChinaThe heritability of milk yield(350 caput), heat tolerance milk yield(350 caput), recta temperature (350 caput) and potassium concentrate oferythrocytes (350 caput) were estimated using half-fellowmen correlationin bull. The results were 0.334, 0.196, 0.20 and 0.318 respectively. Thebreeding value of Israel bulls estimated by BLUP method was 85.12 and itis the highest among the bulls.(2) Study On RAPD Markers And Bioinformatic Analysis Of Heat-Tolerance InHolsteinIn this paper, with DNA from 52 high-milk-yield, strong heat-toleranceand 41 high-milk-yield, weak heat-tolerance Chinese Holstein cattle asmaterial, was studying heat-tolerance in Holstein by using the technologyof PCR-RAPD, with selected 200 RAPD primers,.The results indicated thatthe 581bp 680bp(they are RAPD marker of heat-tolerance inHolstein)specific fragments respectively by using $441 primer and S463primer. Recollected and clone sequenced them, it was found that thesequences of 581bp gene fragment and KCNN2 gene have 90% homology, Ipredicting 581bp gene fragment shoud be intron 3 in KCNN2 gene.Thesequences of 680bp gene fragment and NW001024067.1 have 99%homology. Based on known function of KCNN2 gene, we can deduced KCNN2 geneit is a candidate gene of heat-tolerance. The 581bp(RAPD-s441 marker) genefragment and 680bp(RAPD-S463) gene fragment were submitted to Genbank. Igot GenBank accession numbers EF123743 and EF123744 of the two fragmentsrespectively, In 581bp we did not found ORF, it accord with predicting, In 680bp we found 40RF (ORF1,ORF2,ORF3,ORF4),which are coding 48 aminoacid 34 amino acid 45 amino acid a4 amino acid respectively.(3) Study On SCAR Markers Of Heat-Tolerance In HolsteinBase on sequence of RAPD-S441 and RAPD-S463, used premeier5.0 softwareto design corresponding SCAR premier. The RAPD-S441 marker(581bp) wasconverted to SCAR marker, but RAPD-S463 did not.
