Flax (Linum usitatissimum L.) is an important economic crop for fiber and oil. Amongthe three major bast fiber crops Flax, jute, ramie three, flax has good quality fiber, smallplants, short growing season to seed, self-pollination, small seed and large seed quantities,small genome (2n=30, about 700 Mb), Thus, flax is a model plant for the bast fiber cropsfunctional genomics research. This study was to establish a highly efficient flax regenerationand genetic transformation system for T-DNA insertional mutagenesis and functionalgenomics research. The main results are as follows:1. An efficient plant regeneration system was established through optimization of basicmedium, hormone combination, explants, genotype, seedling age of flax. Data showed thatthe best medium for callus and adventitious shoot differentiation was YB1. Shoot inductionrate of "baihua", "HeiyaNO.4", "K6531", "K7697", "HI026", "I039", "Aliyana" were 98.50%, 98.50 %, 56.50 %, 42.47 %, 54.40 %, 0.00 %, 27.13 %, 97.30 % respectively. Whilethe average shoot numbers per explants were 11.43, 9.33, 2.17, 0.77, 1.10, 0.90, 10.68respectively. Hypocotyls taken from 7 to 10 days old seedlings showed the highestregeneration ability. RB5 was optimum for root formation of regenerated shoot with 100%root induction rate and 15.3 roots per shoot;2. Through the analysis of effects of different antibiotic concentrations, pre-cultivatedtimes of explants, Agro bacterium suspension media, the concentration of Agro bacterium,co-culture temperature and co-culture time on transformation in the experiments, an efficientgenetic transformation system was first established in flax "baihua" variety:3. Transformations were confirmed by Gus stain and PCR. And established a flaxgenetic transformation detection system.
