Strawberry is one of the most important and popular crops around the world forits delicious taste, rich nutrient and economic values. However, productivity andquality are greatly affected by pathogen attack, although improving its resistance hasbeen studied for a long time. It is laborious and slow to create a new cultivar withhigh-multi resistance and high yield and quality through transfer single PR(pathogenrelated )gene. So transferring gene encoding transcription factor into crop plantsmay provide a new approach to improving crop plants with enhanced PR geneexpression and disease resistance. The research using strawberry Fengxiang and Zhangji as materials, theOptimization of regeneration System of strawberry leaf was established. JERF33gene was transferred to Zhangji, then the plant were proved asTransgenic plantsby PCE,DNA dot hybridization. The main results are following:1. A high efficient regeneration system for strawberry leaves was established. The high efficient media for Fengxiangis 1/2MS+2,4-D0.10mg/L +TDZ 4.0mg/L,and for Zhangji is 1/2MS+2,4-D0.07mg/L+TDZ 4.0mg/L, both of the regenerationrate are 100%. The average number of regenerate shoot per leaf for Fengxiang is3.5, for Zhangji is 4.6.2. One week of dark culture is benefit for regeneration of strawberry leaves.3. transfer JERF33 gene plants were proved by PCR and DNA dothybridization. The positive rate are 5.4% and 66.7%4. the result of inoculation of Anthracnose show that ectopic expression ofJERF33 gene in transgenic strawberry plants enhanced the resistance of the plantedto the pathogen.
