Porcine muscle tissue fibroblasts and porcine ear skin fibroblasts in vitro culturewere obtained by tissue pieces.The fibroblast cell lines were establish andcharacterized respectively in this research. The results as follows:1. Morphological observation of the fibroblasts show the shuttle-like shape offibroblasts. Quantity of cytoplasm were large. Karyon were rotundity and situatedmiddle of cytoplasm. The survival rates of fibroblasts were more than 90%.2. Fibroblasts went through lag phase,log phase,plateau phase. The growthcurve of fibroblasts showed "s".3. Tests for microbial contamination from bacteria, fungi were normal,Fibroblasts were not contaminated.4. The morphological observation and RT-PCR and cellular immunochemicalstain used to confirm porcine muscle tissue fibroblasts and porcine ear skin fibroblastsshowed that the purity of cells cultured was more than 90%.5. Karyotype analysis indicated that the porcine muscle tissue fibroblasts andporcine ear skin fibroblasts had 2n=38 chromesomes. The frequency of cellchromosome number to be 2n=38 of porcine muscle tissue fibroblasts was 93.3%; Thefrequency of cell chromosome number to be 2n=38 of porcine ear skin fibroblasts was83.3%.6. Efficiency of cellular transfection were detected by Dual-Luciferase ReporterAssay System. The model of cell culture of porcine muscle tissue fibroblasts andporcine ear skin fibroblasts were successfully established, which has laid goodfoundation for further research on gene function and improving muscle quality.
