Arabidopsis thaliana is regarded as a kind of mode plant,and its effect is more important.people utilize Arabidopsis thaliana in herbage plants but also in wood plants.bamboo is exceptive kind of wood plant,its characteristic is drawn scientist's attention.Apical dominance of bamboo is more blazing than any of other plants.we clone PpTB1 gene from Phyllostachys praecox by dint of OsTB1 and TB1 gene for PpTB1 function.Our lab study designing vectors via bamboo and Arabidopsis thaliana.Ameliorating the methods of Arabidopsis thaliana transformation.The results of study are as follows:1. Constructing express vector.The process that people fulfill plants transformation by dint of express vector, integrating gene to the express vector.We constructed the express vector successfully in this study,and improved the methods of constructing of interim vector. Cnsequently,we can construct express vector efficiently.2. Improving Arabidopsis thaliana transformation.Surfactant is important in the extreme to Arabidopsis thaliana transformation.Types and thickness of surfactant both affect transformation efficiency.In our study,we use a new kind of surfactant Jpx instead of silwet L-77.The new type is equal to the old,even better.Ameliorating environment of incubating Agrobacterium after transformation in 1-2 days,which can enhance transformation efficiency.Our study show,100% air humidity and 28℃temperature is propitious to transformation.
