Ganoderma Lucidum is a kind ofeumycete medicine. It is a big basidiomycetes full of nutrition and high value of health protection, there are kinds of high content of the polysaccharides of the bioactivity active principle in Ganoderma Lucidum, the fruit body polysaccharides, spore power polysaccharides and eumycete polysaccharides generated in the cultivating process of liquid submerged fermentation have the effect of resisting tumor, resisting anti-aging, getting rid of oxygen free radical, promoting blood flow, resisting thrombus and protecting liver. There are also lots of experimental testings for the pharmacologic action of ganoderan peptidoglycan and ganoderan germanium, so more attention has been paid on this. Now the research on Ganoderan mainly are studied on fruit body and spore power, little is studied on submerged fermentation hyphal body and broth. So the process of getting out and freeing polysaccharides from submerged fermentation hyphal body and broth, combining with pharmaco-experiment, has very important meanings to Pharmaprojects.This paper makes research on getting out and freeing polysaccharides from submerged fermentation hyphal body and broth, on antioxygen livenes of different composition, also on how to increase white blood cell of coarse polysaccharides.This research compared antioxygen activity of content of polysaccharides attained bydifferent methods, determining the total content of polysaccharides , using spectrophotometric method and the coloration with phenol-sulphuric acid or anthrone-sulphuric; provilyng some accordance to exact measurement on the content of polysaccharides, according to the analysis on the two method from aspects of the filate , its limitation precision , its stability and its accuracy rating . The result shows that both methods are simple and quick and have also high precesion and good accuracy rating , but the method using phenol-sulphuric requires less complex operation, and the reagent is more stable, there is also not strict controlling required.and phenol-sulphuric acid is the best way to be uusedGetting out of Ganoderma lucidurn hyphal body and broth using hot water, the condensing extract, precipitating different molecular weight of polysaccharides using different dimensional 95% ethanol, after using deproteinization of sevag method , GLP hyphal body, GLPa, GLP19, GLP3tT, GLP47.5, GLP63, GLPT1 and GLP broth will be attained. At the same time , using the 0.5NNaOH to boil the waste, the centrfugating combining filtrate, using third times 95% ethanol to precipitating polysaccharides, GLP and GPLb can be attained after deproteinization using the above method.Analysising antioxygen activity of all the content we had, We got these following results:Ganoderma lucidum byphal body polysaccharides, broth polysaccharides and alkali extracted polysaccharides can clear the OH.generated from the H2O2/Fe2+'s Fenton reaction , and the effect can increase with the more polysaccharides,which means there is a kind of dose-effect relationship between the clearance rate and the polysaccharides; GLP31.7 and GLPf31.7 have highest clearance rate, the clearance rate can reaches about 87.6% at above 200mg/ml, increasing with density increases. The ganoderan has restraint on the generation of O2-, the GLP31.7 can get the highest restraint rate, about 24.3% hyphal body sodas have different effect on the O-2. 's clearance, We can get a conclusion also: All the polysaccharideses attained by different method have disoxidation capability.the higher concentration it is,the stronger its disoxidation capability is under concentration scale between lmg/ml~10mg/ml.and its reducing power and ability of clearing free radicle are basic with one accord, the highest concentration it is,the strongest its disoxidation capability is too.
