| The worldwide-occurred tobacco wildfire is a kind of disease of bacterium that mainlyinfects the tobacco. The physiological differentiation of Pseudomonas syringae pv. tabacicame from different regions in Jilin province was studied firstly with a combine method ofRAPD markers and physiological-biochemical and bacteriocides' sensitivity. The results thatphysiological differentiation of Pseudomonas syringae pv. tabaci were proved by the level ofthe different genes. The results were as following:1 There were 30 strains that came from different regions in Jilin province were tested byFluorescent pigment on KBA, Oxidase, Agrinine dihydrolase, Levan, Patatorot, Nitratereduction, Gelatin liquefaction and Carbon source ultilization.The difference was notsignificant among 30 strains only in Levan there were differences.2 All of the strains' sensitivity to Streptomycin and Streptomycin+ Terramycin weretested by the lowest effective concentration. The results showed that there were significantdifference in sensitivity to Streptomycin between strains came from Huang Nihe in Dun Huaand the others.There were not significant difference in sensitivity of strains with differentregions to Streptomycin+ Terramycin. Streptomycin and Streptomycin+ Terramycin were themost effective bactericide for Pseudomonas syringae pv. tabaci.3 Genome DNA of all strains were analyzed by RAPD with 11 random primers. A total125 RAPD bands were amplified by using 11 random primers, among which,82.4% werefound to be polymorphic.The 30 strains of Pseudomonas syringae pv. tabaci were classifiedseven groups based on cluster analysis(similarity coefficients 0.62). Group A: strains camefrom Huang Nihe in Dun Hua; Group B: strains came from Hei Lin in Yu Shu and two strainscame from Xie Jia in Yu Shu; Group C: one strain came from He Long; Group D: two strainscame from An Tu and Da An; Group E: strains came from Xie Jia in Yu Shu, Da An, Liu He,Ji Dangou in De Hui, Tian Tai in De Hui and Chang Xin in De Hui. Group F: one strain camefrom Liu He; Group G: one strain came from He Long.Group B and Group E were dividedinto more subgroup. Cluster analysis indicated that most of the strains came from the sameareas could be clustered together. The similarity coefficients were from 0.2703 to 0.8431. Theresults showed that the physiological differentiation of Pseudomonas syringae pv. tabaci exists in Jilin province.4 The results by RAPD markers and physiological-biochemical and bacteriocides'sensitivity indicated that the physiological differentiation of Pseudomonas syringae pv. tabaciexists in Liu He, Xie Jia in Yu Shu, Ji Dangou in De Hui and Huang Nihe in Dun Hua.It isfeasible to analyze the physiological differentiation of Pseudomonas syringae pv. tabaci withRAPD markers.5 An optimal PCR system for RAPD in Pseudomonas syringae pv. tabaci was found bythe factors influencing RAPD analysis. |
