Mastitis originates from Greek "mastos" and "suffixitis". Its means mammary gland(person) inflammation. In modern times it refer to mammary gland infection which caused by many kinds of microorganisms. Bovine mastitis is a inflammation in cows mammary gland and often causing serious economic losses in dairy cattle breeding. Bovine mastitis is a complex disease that frequently jeopardize dairy cows. The bovine mastitis whose pathogenic is composed of bacterium, fungus and virus etc., the majority is bacterium. There are over 90ï¼…of the bovine mastitis which caused by bacterium are Staphylococcus aureus, streptococcus and Bacterium coli.The bovine subclinical mastitis has no visible clinical signs, but there is 70ï¼…of the losses caused by it. Thus, the application of ELISA for diagnosis of bovine mastitis will have momentous academic and practice significance. Based on these points, the dissertation would lay stress on the study of bovine subclinical mastitis by double-antibody sandwich ELISA method. It consists of three parts.Part 1 Bovine mastitis, the methods of diagnosis, the immunologic mechanism and the basic principle are summarized in this part. Bovine mastitis is prevalent wordwide, not only causes substantial economic losses to the farming industry but also threaten the health of human potentially. The detection of mastitis usually deployed by clinical, physical and chemical methods. The immunologic mechasim of organism includes physical defence, immunologic mechanism of organism cells and humoral immunity mechanism. The technique of ELISA is more and more perfect as times goes by which can detect antigen and antibody.Part 2 The study and application of double-antibody sandwich ELISA method for diagnosis of bovine subclinical mastitis. Preparated the mouse against bovine and horseradish peroxides conjugated rabbit against bovine immunoglobin G. The study of how to deal with milk and the double-antibody sandwich ELISA was developed. 1050samples of milk from 434 cows were collected and examined by the double-antibody sandwich ELISA. The positive coincidence was 48.9ï¼…. The result in paralleled test ELISA to sodium dodecyl sulfate(SMT) showed that the positive coincidence and negative coincidence were 86ï¼…and 76.9ï¼…respectively.Part 3 Above all, a conclusion was drawn that the double-antibody sandwich ELISA can use as one of the diagnosis for bovine subclinical mastitis because it has a higher sensitivity, specificity and stability.
