| The research was carried out between 2004.9 and 2007.3 in Institute of Forestry and Pomology Beijing Academy of Agriculture and Forestry Science ."Toyonoka","Dudla","Darselect","Tongzi No.1","Tochiotome"were studied as mother ,80,83,94,104,10-5-5 from"Honeoye"which was transferred CBF1 gene were studied as father. The main results were as follows:1 Km of different concentration hardly affected the germination of strawberry seeds, but inhibited the growth of strawberry seedling evidently, inhibiting effect became stronger with the increasing of concentration. Screening medium was MS+Km25mg/L。2 Results of molecular analysis by PCR and southern -blot analysis confirmed that foreign gene CBF1 in transgenic strawberry"Honeoye"could be transferred into cultivar strawberry by crossing.3 16 lines had been obtained and they were 80- Toyonoka,80–Dudla,104- Darselect,94- Darselect.4 Electrolyte leakage tests showed that electrolyte leakage ratio of transferred CBF1 gene plants was lower than control's at every low temperature. Comparaed with cultivar strawberry whose electrolyte leakage ratio rised obviously at-16℃, electrolyte leakage ratio of transferred CBF1 gene plants rised obviously at -20℃.The result showed that the freezing tolerance was increased.5 We used method of freezing survival of plants to test transferred CBF1 gene plant's freezing tolerance, the result showed that 57.7% of the control's and 35.6% of transferred CBF1 gene plants were damaged after treatment of low temperature. 51.9% of the control's and 73.2% of the transferred CBF1 gene plants could grow normally as before. The result showed that the freezing tolerance was increased and CBF1 expressed in the transferred strawberry.6 We used the method of freezing temperature between cells to test plant's freezing tolerance. The freezing temperature between cells of transferred CBF1 plants was -6.11℃,but that of non-transferred CBF1 plants was -4.15℃. Freezing temperature between cells of transferred CBF1 gene plants is 1.96℃lower than control's. |
